Isoform?resolved target discovery and validation in cancer - abstract neosplice therapeutics is developing an isoform-resolved target discovery platform to address one of the most pressing challenges in oncology: The shortage of novel, tumor-specific targets for antibody and t cell- based therapies. Despite major advances in immunotherapy, most patients fail checkpoint inhibitors, underscoring the urgent need for safer, more selective targets. Current therapies rely on a narrow set of well- studied proteins that are also expressed in healthy tissues, leading to on-target/off-tumor toxicities and dose- limiting side effects. Alternative splicing, which occurs in >95% of human genes, creates a vast and underexploited pool of disease-restricted protein isoforms with the potential to serve as highly selective therapeutic targets. The goal of this Phase I SBIR is to establish and validate a pipeline for systematic discovery and validation of tumor-specific isoforms. Aim 1 will use long-read single-cell and bulk RNA sequencing to define isoform repertoires in endometrial cancer (ec) and non-small cell lung cancer (nsclc). Aim 2 will expand these discoveries across large patient cohorts by training a deep learning model (neospliceai) to impute full-length isoform usage from short-read data.
