Remote-controlled pc-car t-cell therapy for hard-to-treat solid tumors - project summary chimeric antigen receptor (car) t-cell therapies have revolutionized blood cancer treatment but remain ineffective in solid tumors due to the scarcity of tumor-specific surface antigens. Many oncogenic drivers reside intracellularly and can only be targeted via peptides presented on major histocompatibility complex (mhc) molecules. However, peptide-mhc complexes are often poorly expressed, and tumors employ immune evasion strategies, limiting antigen availability. This challenge is particularly pronounced in low-mutational burden (tmb) cancers such as neuroblastoma, where traditional car-t and t-cell receptor (tcr)-t therapies struggle. Peptide-centric cars (pc-cars) have emerged as a promising solution, enabling recognition of intracellular tumor antigens via mhc presentation. However, fixed-affinity pc-cars face key challenges, including antigen escape, cross-reactivity, hyperactivation, and exhaustion. To overcome these limitations, mabswitch will develop remote-controlled pc-cars (rpc-cars) targeting a key neuroblastoma oncoprotein using its universal affinity switch for antibodies (unasma) technology.
