Improving the stability of sarna lnp formulations to overcome cold chain challenges - RNA vaccines have transformed the landscape of immunization, enabling rapid design, flexible manufacturing, and strong immunogenicity against emerging pathogens. Their widespread success — most notably during the COVID-19 pandemic — has demonstrated the power of rna-based platforms to respond quickly and effectively to global health threats. However, limitations remain. Rna-based vaccines often require high or repeated dosing to achieve protection which contributes to systemic side effects. Moreover, their inherent thermal instability necessitates stringent cold chain storage and distribution resulting in the wastage of ~50% of doses. These challenges are particularly acute for seasonal vaccines (e.g., influenza) where annual re-dosing is required, and commercial incentives are reduced. To address the challenge of dose/duration of effect, we propose the use of self-amplifying RNA (sarna) — a form of RNA that encodes both the antigen of interest and an alphavirus-derived viral replicase complex, allowing IT to self-replicate within host cells and drive extended antigen expression. This self-driven amplification within the host enables sarna to generate potent immune responses at significantly lower doses than traditional RNA thereby improving tolerability and reducing manufacturing burden.
